Distribución en subpoblaciones peruanas del polimorfismo -13910 C/T en el gen LCT/MCM6 implicado en la persistencia de la lactasa e intolerancia a la lactosa

Abstract
La lactosa es el principal carbohidrato presente en la leche, su malabsorción se debe a la no persistencia de la lactasa, causante de la intolerancia a la lactosa. La no persistencia de la lactasa es variable en las diferentes poblaciones del mundo, y esta condición se relaciona con la variante alélica C del SNP LCT -13910 ubicado en el gen MCM6 y que influye sobre la región promotora del gen de la lactasa. El objetivo de la investigación fue establecer la distribución de las frecuencias del polimorfismo -13910 C/T del gen LCT/MCM6 en diferentes subpoblaciones peruanas. El análisis se realizó con 173 muestras de ADN de las subpoblaciones de Lima (56.1 %), Huarochirí-Lima (13.3%), Puno (17.3%) y Calca-Cusco (13.3%). Se aplicó la metodología PCR-RFLP para evaluar el polimorfismo. Las frecuencias genotípicas en la muestra global fueron: C/C=97,7 % y C/T=2.3%. En las subpoblaciones los resultados fueron: Lima C/C=96.9 % y C/T= 3.31%; en Huarochirí-Lima y en Calca-Cusco el genotipo C/C fue el 100%, en Puno C/C=98.3% y C/T=1,7%. Las frecuencias genotípicas se encontraron en equilibrio de Hardy-Weinberg. En la muestra total, las frecuencias de los alelos fueron: C=98,8% y T=1,2%. Se encontró poca diferenciación genética (índice Fst). En general, no existen diferencias para el polimorfismo LCT/MCM6 -13910 C/T en las subpoblaciones peruanas estudiadas, encontrándose una alta frecuencia de la variante genética C, asociada a la no persistencia de la lactasa e intolerancia a la lactosa.
